Vitamin D Supplementation Showed No Added Pocket Depth Benefit in Meta-Analysis

Vitamin D supplementation did not add a measurable benefit to nonsurgical periodontal therapy in reducing probing pocket depth, according to a systematic review and meta-analysis published Aug. 19 in Evidence-Based Dentistry.

The pooled analysis drew on three studies, covering four subject subsets, that tested supplementation alongside nonsurgical treatment. The estimated mean difference in probing depth was 0.0023 mm, with a 95% confidence interval running from -0.0378 mm to 0.0424 mm, and was not statistically significant at P = .911.

Reviewers searched three databases for randomized controlled trials and controlled clinical trials with at least one month of follow-up, examining changes in probing pocket depth, clinical attachment level, tooth loss, and implant loss. Fourteen articles met the criteria for qualitative assessment: 13 randomized trials and one controlled clinical trial.

None of the 14 examined peri-implant outcomes. The authors identified that absence as a gap in the literature.

The reviewers wrote that vitamin D may exert anti-inflammatory effects on gingival tissue, but that the available evidence is limited, heterogeneous, and insufficient to support a clinical recommendation. They called for longer randomized trials that control for confounding variables.

Only three of the 14 included studies contributed to the pooled probing depth estimate, so the meta-analytic finding rests on a narrow evidence base. The review reports no pooled analysis for clinical attachment level.
